CAS No.: 171596-29-5
Product introduction:Tadalafil is a potent, reversible, and highly selective inhibitor of phosphodiesterase type 5 (PDE5), the enzyme primarily responsible for the degradation of cyclic guanosine monophosphate (cGMP) in the corpus cavernosum. By inhibiting PDE5, tadalafil increases intracellular cGMP levels, leading to smooth muscle relaxation, enhanced blood flow, and maintenance of penile erection in response to sexual stimulation. Unlike other PDE5 inhibitors such as sildenafil (Viagra®) and vardenafil, tadalafil possesses a distinct molecular structure and a significantly longer elimination half-life of approximately 17.5 hours, which earned it the nickname "the weekend pill" for its duration of effect of up to 36 hours. Additionally, tadalafil has been approved for once-daily use at a lower dosage for the treatment of benign prostatic hyperplasia (BPH) and the combination of BPH and erectile dysfunction (ED).

Tadalafil is a white to off-white crystalline powder with the molecular formula C₂₂H₁₉N₃O₄ and a molecular weight of approximately 389.40 g/mol. Its chemical name is (6R,12aR)-6-(1,3-benzodioxol-5-yl)-2-methyl-2,3,6,7,12,12a-hexahydropyrazino[1',2':1,6]pyrido[3,4-b]indole-1,4-dione. The compound is practically insoluble in water but exhibits good solubility in organic solvents such as DMSO (approximately 20 mg/mL) and dimethylformamide. Tadalafil is stable under normal storage conditions when kept in a tightly sealed container, protected from light and moisture, at room temperature. It is typically available in pharmaceutical-grade purity (≥98% by HPLC) for oral formulation into film-coated tablets in various strengths (2.5 mg, 5 mg, 10 mg, and 20 mg).
Core Functions:
Active pharmaceutical ingredient (API) in oral formulations for the treatment of erectile dysfunction (branded as Cialis®) on an as-needed or once-daily basis
Once-daily therapy for the treatment of benign prostatic hyperplasia (BPH) and the combination of BPH and erectile dysfunction (marketed as Cialis® for BPH)
Investigational use in the treatment of pulmonary arterial hypertension, Raynaud's phenomenon, and other conditions involving smooth muscle dysfunction
Research use in pharmacology and urology studies of PDE5 inhibition, cGMP signaling pathways, and vascular smooth muscle physiology
Reference standard for analytical method development and quality control in pharmaceutical testing
