Blonanserin

CAS No.: 132810-10-7
Product introduction:Blonanserin (also known as AD-5423 or Lonasen®) is a novel atypical antipsychotic agent developed by Dainippon Sumitomo Pharma and approved for the treatment of schizophrenia in Japan and Korea. It functions as a potent and relatively selective antagonist at dopamine D₂ and serotonin 5-HT₂A receptors, which are key targets for antipsychotic efficacy . Unlike many first‑generation antipsychotics, blonanserin exhibits a low propensity for extrapyramidal side effects (EPS) and has shown greater efficacy in treating negative symptoms compared to haloperidol . A unique transdermal patch formulation, the first of its kind for an antipsychotic, is also available in Japan, offering an alternative route of administration for patients who have difficulty with oral medication.

Blonanserin is a white to off‑white crystalline powder with the molecular formula C₂₃H₃₀FN₃ and a molecular weight of approximately 367.5 g/mol. Its chemical name is 2-(4-ethyl-1-piperazinyl)-4-(4-fluorophenyl)-5,6,7,8,9,10-hexahydrocycloocta(b)pyridine . The compound is soluble in organic solvents such as DMSO (at concentrations up to 10 mM) and is practically insoluble in water. For optimal stability, blonanserin powder should be stored at -20°C (or 2-8°C for short‑term use), protected from light and moisture, and is typically available in research‑grade purity (≥98% by HPLC).

Core Functions:

  • Active pharmaceutical ingredient (API) in oral formulations for the treatment of schizophrenia, including the management of both positive and negative symptoms

  • Active ingredient in the blonanserin transdermal patch (Lonasen® Tapes) for patients with schizophrenia, particularly those with gastrointestinal dysfunction or difficulty with oral administration

  • Investigational use in palliative care for the management of terminal delirium and refractory dyspnea in terminally ill patients with respiratory diseases

  • Research use in neuropharmacology studies of dopamine D₂ and serotonin 5-HT₂A receptor antagonism, including positron emission tomography (PET) occupancy studies

  • Reference standard for analytical method development and quality control in pharmaceutical testing
